Storing and handling research peptides
A peptide that arrives in good condition can still be compromised on the bench. Most of the loss researchers see — cloudy solutions, drifting results, a vial that no longer behaves like it did last month — traces back to how the material was stored and reconstituted, not to the peptide itself. The good news is that the rules are simple and forgiving once you know them.
Keep the powder cold and dry
Lyophilized (freeze-dried) peptides are the stable form. Kept sealed, dry, and cold, most are stable for extended periods. Two things do the damage: moisture and heat.
- Store the sealed vial at −20 °C or colder for long-term storage. A standard lab freezer is fine for most peptides.
- Let a cold vial warm to room temperature before opening it. Opening a freezer-cold vial invites condensation, and water is the enemy of a dry peptide — especially sequences containing Cys, Met, Trp, or N-terminal Gln.
- Keep desiccant with the material where you can, and reseal promptly.
Reconstitute deliberately
Once in solution, a peptide is far less stable than it was as a powder. Reconstitute only what you need, when you need it.
- Choose a solvent appropriate to the sequence. Sterile water or dilute acetic acid dissolves many peptides; more hydrophobic sequences may need a small amount of a co-solvent. Start gentle.
- Add solvent slowly down the side of the vial and let it dissolve — don’t vortex aggressively if you can avoid it. Foaming and shear aren’t free.
- Bring the vial to room temperature first, and give a stubborn peptide time rather than force.
A useful habit: record the solvent, concentration, and date on the tube the moment you make a stock. Future-you will want to know.
Aliquot to avoid freeze–thaw
Repeated freezing and thawing is one of the most common and avoidable causes of degradation. Each cycle is a small insult; they add up.
The fix is to aliquot once. After reconstituting, split the stock into single-use volumes, then freeze the aliquots. When you need material, thaw one — not the whole batch. You trade a few minutes of pipetting for a stock that behaves the same in week twelve as it did in week one.
Watch for the tells
You don’t need instrumentation to notice trouble. A solution that has gone cloudy, developed a precipitate, or changed color deserves suspicion. So does a result that has quietly drifted across a series of experiments using the “same” stock. When in doubt, make a fresh aliquot from frozen powder and compare.
The short version
Cold, dry, and dark for the powder; room-temperature before opening; gentle reconstitution; and single-use aliquots so nothing gets thawed twice. None of it is exotic — it’s just the discipline that keeps a good material good.
